Increased Antimutagenic and in Vitro Anticancer Effects by Adding Isoflavone and Mistletoe Extracts during Doenjang Fermentation

Abstract
Antimutagenic and in vitro anticancer effects of doenjangs that added with isoflavone and mistletoe extracts and using bamboo salt were studied by Ames test using Salmonella typhimurium TA100 and 3-(4,5- dimethylthiazol)-2,5-diphenyltetrazolium bromide (MTT) assay using PC-3 and DU145 human prostate cancer cells, respectively. In the antimutagenicity test, 1.1% isoflavone extract added doenjang exhibited 81% antimutagenicity against aflatoxin B1 (AFB1), at the 1.25 mg/plate concentration while the control doenjang revealed 63% antimutagenicity, showing the increased antimutagenic effect by the addition of isoflavone extract during the doenjang fermentation. Isoflavone extract added doenjang also increased antimutagenic effect against N-methyl-N¡¯-nitro-N-nitrosoguanidine (MNNG). The inhibition rate
of the control doenjang showed 34% at 0.625 mg/plate, while 0.55% isoflavone extract added doenjang inhibited by 65% and 85% at the 0.625 and 1.25 mg/plate, respectively (p£¼0.05). Mistletoe extract added doenjang showed 66¡­90% and 57¡­80% inhibition rates against isoflavone mutagenesis induced by AFB1 and MNNG, respectively. In MTT assay, isoflavone extract added doenjangs caused 71¡­74% inhibition on the proliferation of PC-3 human prostate cancer cells at 0.5 mg/ml while the control doenjang exhibited only 46% inhibition. And mistletoe extract added doenjangs showed 56¡­79%
anticancer effects on PC-3 and DU145 human prostate cancer cells at 0.5¡­1.0 mg/ml. From these results, it can be concluded that isoflavone and mistltoe extract addition to the doenjang during the preparation increased the antimutagenic and in vitro anticancer activities of the doenjang. Bamboo salt instead of natural salt used doenjangs increased antimutagenic and in vitro anticancer activities at the same treatment concentration (0.55%). The activities were same or even more significant (p£¼0.05) compared to the results at the same or double concentration. (Cancer Prev Res 12, 68-74, 2007)
